Résumé

The effect of culture medium formulation and supplementation on human chorionic gonadotropin (hCG) prodn. in primary culture of villous trophoblast was studied. Two culture media and six supplementations were analyzed. Cells were seeded in and grown in DMEM-High Glucose or M199 supplemented in the presence or absence of different sera or other supplements: 10% Hyclone fetal bovine serum (FBS), 10% Sigma FBS, 10% charcoal/dextran (C/D) treated Hyclone FBS, 0.2% Hyclone FBS or 1X SITE+3. Results demonstrated that media formulation and supplementation does have an effect on hCG release (biochem. differentiation) and protein content (cell viability) in primary culture of villous trophoblast.
