Characterization of the 5-HT2A serotoninergic receptor in human placental choriocarcinoma cells lines

Résumé


We have recently demonstrated a mitogenic effect of serotonin via stimulation of 5-HT2A receptors in placental choriocarcinoma cell lines, JEG-3 and BeWo. The mechanism through which the 5-HT2A receptors perform this stimulating effect on placental cell proliferation is still unknown.

Objectives and methods: In the present study, we investigated, by co-immunoprecipitation and western blot analysis, the implication of 1) the Gq/11-PLC-β-MAP kinase and 2) the JAK/STAT signaling pathways following the activation of placental 5-HT2A receptors.

Results: Our results showed that stimulation of the choriocarcinoma cell lines, JEG3 and BeWo, by a specifi c 5-HT2A receptor agonist, DOI, leads to a JAK2 and 5-HT2A interaction as well as to a increase of the interaction between Gq/11 and 5-HT2A receptor. Moreover, our data demonstrated that the activation of the 5-HT2A receptor in these cell lines activates both the MEK-ERK1/2 and JAK2-STAT3 signaling pathways.

Conclusion: These results shown that the activation of the 5-HT2A receptors stimulate the JAK2/STAT3 pathway and Erkl/2 pathway in the JEG-3 and BeWo cell lines. More studies are necessary to fi nd out which pathway or pathways are responsible for the increase of cell proliferation by 5-HT.
