Modulation of placental serotonin transporter and 5-HT2A-receptor by gestational diabetes mellitus

Résumé


Objectives: The presence of 5-HT2A-serotonin receptor (5-HT2AR) and serotonin transporter (SERT or 5-HTT) has been demonstrated in human term placental tissue as well as in choriocarcinoma cell lines. However, the role of the serotonin system in regulation of placental function and pregnancy remains unclear. Recent studies reveal that serotonin might be implicated in the etiology of gestational diabetes mellitus (GDM). Therefore, the aim of this study was to determine which type of cells express SERT and 5- HT2AR in the normal human term placenta and investigate the effect of GDM on the expression of both placental SERT and 5-HT2AR.

Methods and Results: Immunohistochemical analysis shows that both SERT and 5- HT2AR are present in the villous trophoblast and the fetal capillary endothelium of normal term placenta. The expression of SERT and 5-HT2AR in the villous cytotrophoblast and syncytiotrophoblast cells primary culture was confirmed by quantitative western blot and RT-PCR analysis. A significantly decreased expression of SERT (56.3 % mRNA and 76.7 % protein) and 5-HT2AR (79.1 % mRNA and 29.1 % proteins) was observed in placental tissues from GDM compared to non-GDM pregnancies.

Conclusion: This data suggests that SERT and 5-HT2AR might be implicated in the etiology of GDM. Moreover, the presence of SERT and 5-HT2AR in villous trophoblast cells argue in favor of an important role of serotonin in human placental function. Further studies are needed in order to elucidate the role of the placental serotonergic system and its implications in normal pregnancies and those with GDM complications.
