Résumé

L’ARN est capable de se replier en structures secondaires et tertiaires par la formation des paires de base canoniques et non canoniques et ce repliement est important pour le bon fonctionnement des ARN régulateurs. Cela représente un défi pour les logiciels bioinformatiques qui font la conception des ARN synthétiques qui doivent conserver la structure et fonction de l’ARN original. Dans ce projet nous avons utilisé deux nouveaux outils bio-informatiques ‘Ribosoft’ et ‘Enzymer’. L’un fait la conception des ribozymes en tête de marteau et l’autre permet la génération de nouveaux ARN de n’importe quel type. Le ribozyme en tête de marteau est un ARN noncodant auto-clivant actif en cis et en jouant sur sa séquence il peut être actif en trans pour cliver d’autres ARN d’une façon spécifique, il était utilisé dans ce projet pour développer un circuit logique qui permet le contrôle de l’expression d’un gène désiré. Le circuit est formé par des ribozymes sous le contrôle de riboswitchs. Les riboswitchs sont des ARN noncodants agissant comme des récepteurs capables de contrôler l’expression d’un gène situé en aval et leur activité est dépendante de la présence ou l’absence d’un ligand spécifique. Le rôle du riboswitch dans le circuit logique est le contrôle, via son ligand, de l’expression du ribozyme qui contrôle l’expression du gène cible.

RNA can fold into secondary and tertiary structures by the formation of canonical and non-canonical base pairs. This folding is important for the proper functioning of regulatory RNAs. This represents a challenge for bioinformatic softwares for the design of synthetic RNA that must maintain the structure and function of the original RNA. In this project, we used two new bioinformatics tools 'Ribosoft' and 'Enzymer', the first can design new hammerhead ribozymes and the other allows the generation of new RNAs of any type. The hammerhead ribozyme is a non-coding RNA with a self-cleaving activity in cis, but by modifying its sequence it can be made to act in Trans to cleave other RNAs in a specific way. The ribozyme was used in this project to develop a logic circuit that enables the control of the expression of a desired gene. The circuit is formed by riboswitches in addition to ribozymes. Riboswitches are allosteric non-coding RNAs capable of controlling the expression of a gene located upstream of them and their activity is dependent on the presence or absence of a specific ligand. The role of the riboswitch in the logic circuit is the control, via its ligand, of the expression of the ribozyme that controls the expression of the target gene.
